DONSON: a critical regulator of DNA replication and genome instability
Abstract:
DONSON is a key component of the replisome that is not only required to initiate DNA replication but also stabilise and protect stalled replication forks during genome duplication, in part through its ability to facilitate activation of ATR. As a consequence, compromising DONSON function results in highly elevated levels of spontaneous replication stress and genome instability, which in humans, causes severe neurodevelopmental abnormalities, such as microcephaly and growth retardation. Whilst DONSON is essential for normal DNA replication, very little is known about how it functions to stabilise damaged replication. During this talk, I will present new evidence about how post-translational modification of regulation of DONSON is critical for its ability to protect and stabilise damaged replication forks
