In the past ten years, Prof Carr has focused his laboratory on understanding the link between replication and chromosomal rearrangements (CRs). His laboratory developed a site-specific replication fork barrier system in fission yeast and used this to demonstrate for the first time a direct link between replication restart and homologous recombination dependent CRs. His laboratory has subsequently built on this experimental system to: demonstrating that recombination restarts replication without a DSB intermediate; uncover that restarted replication forks are non-canonical and error prone; and delineate previously unknown pathways for the generation of acentric and dicentric chromosomes -the precursors of many oncogenic CRs.
